    Intérêts de recherche

    Depuis les dix dernières années, mes recherches sont consacrées à la validation de l'hypothèse voulant qu'un déséquilibre oxydant-antioxydant chez le nouveau-né est à l'origine de nombreuses complications néonatales dont l'incidence et la gravité de certaines dépendent du genre. Le déficit de la capacité antioxydante du nouveau-né humain a été observé lors d'études enzymologiques démontrant une faible concentration tissulaire en glutathion (molécule clé de la défense antioxydante) chez le prématuré et davantage chez le garçon.

    Ce faible taux de glutathion n'est pas lié à la capacité de synthèse, ni à sa régénération mais bien à l'immaturité du transport transcellulaire de cystéine (substrat pour la synthèse de glutathion). D'un autre côté, la nutrition parentérale constitue une charge importante d'oxydants. L'exposition des multivitamines parentérales à la lumière ambiante est la source principale de peroxydes dans la nutrition parentérale. Leur génération est due à l'interaction entre l'oxygène dissout et l'acide ascorbique, le tout catalysé par la riboflavine activée par la lumière. L'infusion d'une nutrition parentérale exposée à la lumière induit chez l'animal une fibrose pulmonaire (causée par les peroxydes), une faible alvéolarisation (causée par un produit de photodégradation de la vitamine C), une stéatose hépatique ainsi qu'une hypertriglycéridémie. Nous avons démontré que, chez l'enfant, la photoprotection de la nutrition parentérale prévenait l'élévation des peroxydes urinaires et l'hypertriglycéridémie. Par spectrométrie de masse, nous avons également trouvé une nouvelle voie de dégradation de la vitamine C conduisant à la formation d'un ascorbylperoxyde. Chez l'animal, la concentration urinaire de cette nouvelle molécule est corrélée avec l'activité de l'acétyl CoA carboxylase hépatique (enzyme clé de la synthèse des acides gras).
